Using the cultural-ecological theoretical framework and the Stages of Migration framework, this study utilized an existing data set to explore the relationships between migration stress, psychological distress, experiences of discrimination, and awareness of discrimination in relation to educational outcomes in a sample of 281 Latino immigrant youth. These relationships were then examined to see if they differed as a function of perceived support, gender, and school type (i.e., middle school versus high school). Structural equation modeling was utilized to test the hypothesized model that included migration stress, psychological distress, and education outcomes. The structural model showed very good fit. Results suggest that migration stress has a significant direct effect on psychological distress and on educational outcomes among Latino immigrant youth. Participants reporting high migration stress reported greater psychological distress and had poorer educational outcomes with respect to academic grades, educational aspirations, and educational expectations. Moderation testing indicated the structural model did not vary as a function of perceived support, gender, or school. Implications for research and practice are discussed.   Citation × Citation Citation Abstract Copy Title: High-Resolution Studies Link Vascular Endothelial Growth Factor Signaling with Endocardial-Myocardial Dynamics Controlling Heart Ventricle Development Author: Karfilis, Kate Year: 2016 Determining how coordinated gene expression changes direct embryonic heart development is paramount to understanding the genetic causes and developmental origins of congenital cardiomyopathies. Towards this goal, I present an optimized protocol for mouse thiouracil tagging (TU-tagging), a novel transcriptomics methodology for defining dynamic and cell specific gene expression programs, and validate TU-tagging for cardiovascular research. I then apply related and additional high-resolution approaches to characterize how vascular endothelial growth factor (VEGF) signaling coordinates cell and gene expression dynamics underlying heart ventricle development. TU-tagging is a powerful approach to study dynamic gene expression programs of defined cell types while they are natively embedded within a complex organ. TU-tagging integrates genetic and chemical approaches to provide temporally controlled in vivo covalent labeling of cell type–specific RNA. Here, I describe two significant optimizations of the TU-tagging molecular biology and bioinformatics workflows that improve the method’s ability to identify differentially expressed genes and expand the technology’s utility. Next, using chemical inhibition of VEGF signaling in combination with high-resolution imaging and transcriptomic methods, I show that VEGF signaling directly promotes formation of the trabeculae that uniquely develop in the heart ventricle. By RNA-Seq, I identify VEGF-dependent target genes, including Gpr126 and Bmp10, which encode additional signaling proteins. I further show that myocardial Bmp10 expression and resulting endocardial Bmp10-driven pSMAD1/5/8 signaling is under sustained control by endocardial VEGF signaling. This continuous VEGF-BMP signaling interplay between endocardial and myocardial cells led me to examine the dynamic tissue arrangements between the two cell types during early stages of trabeculation. By extensive staining and high resolution imaging, I show that endocardial cells can be subdivided into three classes; 1) quiescent cavity cells that are well-separated from the myocardium, 2) proliferative and signal responsive transition zone/stalk cells that directly interact with myocardium to coordinate both cell types’ activities, and 3) CD34-expressing migratory tip-like cells uniquely found at the base of forming trabeculae. VEGF promotes trabeculation by 1) driving proliferation of endocardial transition zone/stalk cells and, secondarily, neighboring myocardium, and 2) directing the outward migration of endocardial tip cells that causes myocardial tissue to accumulate within individual and extensive ridge-like trabeculae. Defining these multiple roles of VEGF signaling during ventricle development reveals a novel conceptual framework for understanding trabeculation mechanisms and therefore processes likely to be disrupted in common congenital cardiomyopathies. While the predominant historiography of these migrations has focused on a binary of hostile or non-hostile interactions between Indians and emigrants, the focus on violence has obscured the larger issue of frequent and amicable interactions between emigrants and Indian peoples along the overland route. Factors such as trade, the availability of resources, and cultural differences influenced the nature of these inter-ethnic interactions, which varied from the beginning of the trail on the Plains to the end of the trail on the Columbia Plateau. Autopsie in vivo de quelques romans d’auteures caribéennes et subsahariennes (Condé, Mukasonga, Danticat et Miano)” examines the complex logics examines the complex logics of social exclusion and connects writings from Sub-Saharan Africa and the Caribbean, two sites often treated separately in the domain of Francophone studies. Precisely, this dissertation addresses how exclusion unfolds in these postcolonial societies—with migration, exile, and globalization echoed in the literary texts that I read. My argument is that our understanding of social exclusion and violence in these societies still draws solely from homogenizing development theories that originate outside of them. Re-theorizing social exclusion, I show in my work how these texts portray acts of social exclusion and violence through such insidious categories as geography, origins and lineage, as well as personal history, and local traditions and practices, that contribute to the making of misfits and outcasts, and yet remain overlooked in most attempts to address social exclusion in these specific locations. In navigating these relationships between social situations and literary form, I engage with psychology, social theory, and also physiology as I resort to autophagy (from the Greek “auto” meaning self and “phagy” meaning eating), a physiological process in the body that destroys cells to analogically demonstrate that by nurturing destructive behaviors these societies jeopardize chances to reach national cohesion and therefore contribute to their own destruction. The various chapters analyze texts by women writers: French Guadeloupian Maryse Condé, Haitian-American Edwidge Danticat, Rwandan French Scholastique Mukasonga, and Cameroonian French Léonora Miano. Self-critical agents of their communities, their act of bearing witness to these disruptions from a decentered position becomes highly problematic specifically for Danticat and Miano, as their legitimacy is challenged by resisting readers from their countries of origin who see their hyphenated selves as outsiders and traitors. But, hardly discouraged, these authors demonstrate the need for a renewed social response in writing that is provocative, with a rhetoric that resists the obsolete framing of fault and responsibility as always the Other’s.
