Estimating the true CO2 in melt inclusions is difficult, as much is lost to shrinkage bubbles, which form upon post-entrapment cooling and crystallization. Reheating olivine to temperatures above the melt inclusion trapping temperature and then quenching rapidly can restore CO2 to the glass because the CO2 in the bubble redissolves at high temperature. Previous work has established that olivine crystallization for the1959 Kilauea Iki eruption took place in the shallow summit reservoir, but crystallization depths have not been established for the rift extension of the eruption, at Kapoho. The new data presented here suggest that the most primitive Kilauea Iki component bypassed the summit reservoir for the east rift zone prior to the start of the eruption and was later erupted at Kapoho. Understanding the effects of climate change on AMF, and the resulting plant responses, is a crucial factor in predicting ecosystem responses on a global scale. We used a manipulative climate change experiment embedded within a natural climate gradient in Oregon and Washington to examine how the effects of future climate change on AMF-plant symbioses are mediated by soil water availability, soil nutrient availability, and vegetation dynamics. Using structural equation modeling, we found that the direct effect of increasing temperatures was to decrease AMF colonization. Indirect effects of temperature, mediated through other variables, canceled each other out. However, future shifts in these relationships could either exacerbate or mitigate the negative direct effect of temperature. As ecosystems in Mediterranean climates experience more intense droughts and heavier rains, decreases in AMF colonization could have substantial consequences for plant communities and ecosystem function.   Citation × Citation Citation Abstract Copy Title: Theoretical Study of Spin-wave Effects in Quantum Ferromagnets Author: Bharadwaj, Sripoorna Paniyadi Krishna Year: 2017 In this dissertation, we examine quantum ferromagnets and determine various effects of the magnetic Goldstone modes or "magnons'' in these systems. Firstly, we calculate the magnon contribution to the transport relaxation rate of conduction electrons in metallic ferromagnets and find that at asymptotically low temperatures, the contribution behaves as T^2 exp(-T_0/T) and not as T^2 predicted previously. To perform these calculations, we derive and use a very general effective theory for metallic ferromagnets. This activation barrier-like behavior is due to the fact that spin waves only couple electrons from different Stoner subbands that arise from the splitting of the conduction band in presence of a nonzero magnetization. The T^2 behavior is found to be valid only in a pre-asymptotic temperature window. The temperature scale T_0 is the energy of the least energetic ferromagnon that couples electrons of different spins. Second, we discuss magnon-induced long-range correlation functions in quantum magnets. In the ordered phases of both classical ferromagnets and antiferromagnets, the long-range correlations induced by the magnons lead to a singular wavenumber dependence of the longitudinal order-parameter susceptibility in spatial dimensions 2<d<4. We investigate the quantum analog of this singularity using a nonlinear sigma model. In a quantum antiferromagnet at $T=0$, a weaker nonanalytic behavior is obtained, which is consistent with power counting. The analogous result for a quantum ferromagnet is absent if the magnon damping is neglected. This is due to the lack of magnon number fluctuations in the quantum ferromagnetic ground state. Magnon damping due to quenched disorder restores the expected nonanalyticity. Finally, we use an effective field theory for clean, strongly interacting electron systems to calculate the magnon contribution to the density of states, the longitudinal magnetic susceptibility and the conductivity in an itinerant ferromagnet. Utilizing a loop expansion that does not assume the electron-electron interaction to be a small parameter, we obtain the leading nonanalytic corrections to the Stoner saddle-point results for these observables, as functions of the frequency and wavenumber in the hydrodynamic limit. Many variables influence timescales of equilibration between vapor and melt after a perturbation in pressure, temperature, or other factors, and the magnitude of this equilibration time determines whether the system experiences equilibrium degassing or not. In order to create a simplified framework for assessing degassing regime, we constructed a numerical diffusion model to test the sensitivity of equilibration time to variables such as bubble size, spacing, melt temperature, initial and final system pressures, and water content. We then determined the degassing regime for a range of bubble-spacing and decompression rates as an initial simplified framework to build on. We also attempted the first mixed-volatile continuous decompression experiments in order validate our model and further improve analyses and interpretations of volatile gradients in natural samples. In part 2, I applied the equilibrium terrestrial biosphere models, BIOME4 and BIOME5 (developed from BIOME4 herein), for reconstructing paleoclimate. I applied inverse modeling through the iterative forward-modeling (IMIFM) approach that uses the North American vegetation data to infer the mid-Holocene (MH, 6000 years ago) and the Last Glacial Maximum (LGM, 21,000 years ago) climates that control vegetation distributions. The IMIFM approach has the potential to provide more accurate quantitative climate estimates from pollen records than statistical approaches. Reconstructed North American MH and LGM climate anomaly patterns are coherent and consistent between variables and between BIOME4 and BIOME5, and these patterns are also consistent with previous data synthesis. Progress is being made towards combating the leading cause of unavoidable blindness, age-related macular degeneration (AMD). AMD affects ten million Americans and costs the world economy $343 billion annually. Retinal implants promise to restore sight by replacing the eye's damaged photoreceptors with electronic photodiodes. Clinical trials succeed at restoring some vision, but are limited by the stimulating electrodes. We study the electrode-neuron interface with a focus on the geometrical dependence of the electrode. The functionality of neurons is intimately connected to their branching and curving shape, described by fractal geometry. We examine the morphology of neurons using fractal analysis. The results inform our electrode designs, which are fabricated using top-down lithographic and bottom-up self-assembly techniques. A novel technique for fabricating a fractal electrode is presented. Heating and cooling a film of poly(methyl methacrylate) on a SiO2 substrate causes fractal structures to form on the surface. The geometry of the structures is temperature dependent, producing crystalline branches at lower temperatures and diffusion-limited aggregates at higher temperatures. Subsequent deposition of antimony nanoclusters shows preferred diffusion to the fractal surface features. The dependence of a photodiode's performance on its top contact geometry is explored using modified nodal analysis. The results reinforce the need to balance a low mean semiconductor-metal separation distance with an adequate contact width for low resistance, all while maximizing light input. Future designs will benefit from the spatial voltage maps produced by the simulation. The electric field emanating from an electrode is also dependent on the geometry of the electrode. The Faraday cage effect is exploited to achieve similar electric field responses to traditional electrode shapes. A preliminary study of neural adhesion to SU-8 fractal electrodes is promising. The neuron grows along the electrode even at 90° turns. The role the fractal geometry plays in neuron and electrode functionality is shown to be significant. Progress is being made towards combating the leading cause of unavoidable blindness, age-related macular degeneration (AMD). AMD affects ten million Americans and costs the world economy $343 billion annually. Retinal implants promise to restore sight by replacing the eye's damaged photoreceptors with electronic photodiodes. Clinical trials succeed at restoring some vision, but are limited by the stimulating electrodes. We study the electrode-neuron interface with a focus on the geometrical dependence of the electrode. The functionality of neurons is intimately connected to their branching and curving shape, described by fractal geometry. We examine the morphology of neurons using fractal analysis. The results inform our electrode designs, which are fabricated using top-down lithographic and bottom-up self-assembly techniques. A novel technique for fabricating a fractal electrode is presented. Heating and cooling a film of poly(methyl methacrylate) on a SiO2 substrate causes fractal structures to form on the surface. The geometry of the structures is temperature dependent, producing crystalline branches at lower temperatures and diffusion-limited aggregates at higher temperatures. Subsequent deposition of antimony nanoclusters shows preferred diffusion to the fractal surface features. The dependence of a photodiode's performance on its top contact geometry is explored using modified nodal analysis. The results reinforce the need to balance a low mean semiconductor-metal separation distance with an adequate contact width for low resistance, all while maximizing light input. Future designs will benefit from the spatial voltage maps produced by the simulation. The electric field emanating from an electrode is also dependent on the geometry of the electrode. The Faraday cage effect is exploited to achieve similar electric field responses to traditional electrode shapes. A preliminary study of neural adhesion to SU-8 fractal electrodes is promising. The neuron grows along the electrode even at 90° turns. The role the fractal geometry plays in neuron and electrode functionality is shown to be significant. Continued study of, and experimentation with, new electrode designs is sure to produce exciting possibilities in the future. This dissertation includes previously unpublished co-authored material. Several TSe2, [(SnSe)1+y]m(TSe2)n, [(SnSe)1+y]m(TSe2)n[(SnSe)1+y]p(T'Se2)q, and (SnSe)1+y(V1-xTaxSe2) layered compounds were synthesized by the new modulated elemental reactant (MER) technique with T = V, Ta, and Ti. The MER approach is a low-temperature synthesis that allows the kinetic trapping of metastable compounds, allowing a designed synthesis with control over the value of m, n, p, and q. These layered compounds were structurally characterized by X-ray diffraction and transmission electron microscopy studies. Each integer increase of m, n, p, and q resulted in a linear increase in thickness, representing the single structural units of SnSe (a Sn/Se atomic bilayer) and TSe2 (an X-T-X trilayer sandwich). All of the compounds contained preferentially oriented layering, with the layer's (00l) plane parallel to the substrate surface. From Bragg-Brentano and in-plane X-ray diffraction studies, the layers are found to be regularly spaced along c with abrupt interfaces and contain crystallinity in their ab-plane. Many of these layered compounds were found to exhibit turbostratic disorder, a random rotational disorder that is usually present in materials made by the MER technique. The presence of turbostratic disorder was found to be dependent on the polytypes that exist in the bulk form of the TSe2 constituent. The electrical properties of the layered compounds were studied by means of temperature-dependent resistivity and Hall measurements. Interesting electrical properties were found as a result of the turbostratic disorder, including a charge density wave transition found in the [(SnSe)1.15]m(VSe2) ferecrystal. The onset temperature of the CDW transition was found to be a sensitive function of the layering sequence, increasing with higher m values. The CDW transition was attributed to the VSe2 constituent and was found in all the composite crystals that contained VSe2. As a programmable material, our metafluid represents a new platform on which to study complex phenomena such as self-assembly and pattern formation. We demonstrate one such application in our study of short-chain polymer analogs embedded in our system. Gametogenesis and spawning occur seasonally from May through September, when water temperatures exceed 14.5&deg C, with brooding oysters found from July through September. Oocyte diameters increased significantly from May to June, and from June to July within oyster populations at Haynes Inlet and Coalbank Slough, respectively. Male gametogenesis initiated in May at Haynes Inlet and in June at Coalbank Slough. Dry meat condition values increased significantly during periods of reproduction and decreased following the reproductive season's end. Condition index values for Coalbank Slough were consistently lower than those at Haynes Inlet, suggesting poor nutrition or physiological stress. Salinities below recorded physiological thresholds are believed to be the primary environmental factor influencing the discrepancy in reproductive activity at Coalbank Slough. I combine extensive data on aid received by Nicaraguan households with data on municipal election outcomes and an exogenous, precipitation-based measure of hurricane impact. I find that the hurricane had long-lasting effects on the aid received by households from both NGOs and the government. In the short term, however, the government did not provide aid according to the objective measure of hurricane damage but instead provided aid along political lines. The second essay presents estimates of a relationship between extreme hot temperatures during gestation and a child's subsequent physical well-being in a sample of children in Peru, thus extending existing evidence constructed from U.S. data. Estimates are constructed using high-resolution gridded climate data and geo-coded household surveys. The results suggest that a period of extreme heat (a month whose average temperature is more than 2 standard deviations above the local average) in the period 1 to 3 months before birth is associated with lower weight at birth and a reduction in height (measured 1 to 59 months after birth) that cannot be fully explained by birth weight. There is no evidence of differential maternal investment, as measured by duration of breastfeeding, according to a child's exposure to extreme heat during gestation. The third essay asks whether improved treatment of HIV/AIDS in Africa can be achieved simply by paying health workers to do more. I present estimates of the impact of financial incentives paid to individual workers at public health facilities in Mozambique. The results suggest that piece-rate incentives increased the delivery of five out of fourteen health services for which treatment effects can be identified, with estimated increases ranging from 34 to 157 percent, depending on the particular service. I find no evidence of a corresponding decrease in the delivery of services that are not financially incentivized, suggesting that there is no "crowding out" of intrinsic motivation. Geomorphic factors, such as channel geometry, planform, and sediment transport, are major determinants of the value of river systems, including their suitability for threatened and endangered species and for human uses of water. This dissertation research uses a hierarchical modeling approach to investigate potential impacts of anthropogenic climate change on river morphology in the interior Pacific Northwest. The research will address the following theoretical and methodological objectives: 1) Develop downscaled climate change scenarios, based on regional climate-model output, including changes in daily minimum and maximum temperature and precipitation. 2) Estimate how climate change scenarios affect river discharge and suspended-sediment load, using a basin-scale hydrologic model. 3) Examine potential impacts of climate-driven hydrologic changes on stream power and shear stress, bedload sediment transport, and river morphology, including channel geometry and planform. The downscaling approach, based on empirically-estimated local topographic lapse rates, produces high-resolution climate grids with positive forecast skill. The hydrologic modeling results indicate that projected climate change in the study rivers will change the annual cycle of hydrology, with increased winter discharge, a decrease in the magnitude of the spring snowmelt peak, and decreased summer discharge. Geomorphic modeling results suggest that changes in reach-averaged bedload transport are highly sensitive to likely changes in the recurrence interval of the critical discharge needed to mobilize bed sediments. This dissertation research makes an original contribution to the climate-change impacts literature by linking Earth processes across a wide range of spatial scales to project changes in river systems that may be significant for management of these systems for societal and ecological benefits. Passive heat stress results in elevations in core temperature (inducing heat shock protein expression) and changes in cardiovascular hemodynamics, such as increased cardiac output and shear stress, that are similar to exercise. Thus, repeated passive heat stress (“heat therapy”) may provide an alternative means of improving cardiovascular health, particularly for patients with limited exercise tolerance and/or capabilities. Therefore, the goal of this dissertation was to perform integrative studies to determine the effects of heat therapy on vascular function and the associated cellular pathways in young, sedentary humans. Twenty subjects were assigned to participate in 8 weeks (4-5x/week) of heat therapy (N=10; immersion in a 40.5°C bath sufficient to maintain rectal temperature ≥38.5°C for 60 min/session) or thermoneutral water immersion (N=10; sham). As discussed in Chapter V, we found that heat therapy improved numerous well-established biomarkers of conduit vessel/macrovascular function, including flow-mediated dilation (a measure of endothelial function), arterial stiffness, intima media thickness, and blood pressure. Heat therapy also improved microvascular function, as discussed in Chapter VI, measured as improved cutaneous thermal hyperemia and nitric oxide-dependent dilation (the difference between microdialysis sites receiving Lactated Ringer’s [control] and nitric oxide synthase inhibition). No changes were observed in any variables in sham subjects. In Chapter VII, we showed that both direct cellular heating and serum collected from human subjects following heat therapy improved nitric oxide bioavailability and angiogenesis in cultured endothelial cells, providing potential mechanisms by which heat therapy improves vascular function in vivo. Therefore, the studies described herein provide comprehensive evidence that passive heat therapy improves vascular health and insight into the mechanisms involved. Our data presented in Chapters IV-VII, combined with pilot data we conducted in spinal cord injured individuals (Chapter VIII), strongly indicate that passive heat therapy could be used as a simple and effective tool to improve cardiovascular health in a variety of patient populations. Such bidentate ligands, however, often suffer from synthetic challenges, which can be alleviated by the use of simpler monodentate ligands that assemble through non-covalent interactions to mimic the structure of bidentate ligands at the metal center. To produce a strongly assembled catalyst complex, the Hamilton receptor motif was utilized. Hamilton receptors form six hydrogen bonds with complementary guests and have binding affinities for barbiturates of up to 104 M-1 in CDCl3. Complete bifurcation of the Hamilton scaffold produces a modular ligand structure that allows for modification of either end of the supramolecular ligand structure. Similarly, the barbiturate guest can be synthetically altered creating both chiral guests and guests with differing amounts of steric bulk. Both experimental titration data and density functional theory calculations show that steric bulk discourages binding of the guest while a pre-organized host encourages guest inclusion. Electronic effects on the bifurcated Hamilton system were studied by varying the electron donating or withdrawing ability of the benzamide moiety on the host molecule. Electron withdrawing moieties produce more acidic amide hydrogens on the host which are able to participate in stronger hydrogen bonds with the guest resulting in a stronger host-guest complex. The effects of substitutions on the barbiturate guest were examined as well, and increased steric bulk on the guest resulted in decreased affinities with the host. The bifurcated Hamilton receptor ligands were examined in the palladium-catalyzed Heck reaction of iodobenzene with butyl acrylate. Pd2(OAc)4 was used as a control and all reaction yields with the diphenylphosphine ligand-stabilized Pd were greater than or equal to those obtained with Pd2(OAc)4 alone. The reaction rates did not correlate with the determined binding constants, suggesting that phosphine substitution on the guest plays a larger role than affinity of the complex for the guest. Reaction temperatures were varied, and at lower temperatures the yields increased implying that the strength of the hydrogen bonds between the metal complex and the guest does play a secondary role in the catalysis. The compounds are prepared from modulated elemental reactants and display interesting structural and electronic behavior. Section 1 of this dissertation outlines initial attempts to characterize constituent interaction. The first member of the SnSe based subclass is reported and displays the highest Seebeck coefficient of any m = n = 1 compound reported to date, and a surprising amount of order is observed, compared to previously reported compounds. With properly established deposition parameters, the synthesis was extended to included the m = 2-4 compounds. These compounds display interesting electronic behavior that suggests the band structure shifts considerably as the SnSe block is expanded, affecting the interaction between the constituent layers. The first compound based on BiSe is then reported, suggesting that the Bi structure donates more conduction electrons to the band structure. Targeted substitution through kinetic control is the focus of Section 2, and a family of (PbxSn1-xSe)1+δTiSe2 is reported over the entire range of x, even though a miscibility gap exists in the bulk PbxSn1-xSe system. The resulting alloyed intergrowth compounds also display equal or higher mobility than the end members, suggesting modulation doping could be used to affect transport properties. As a proof of principle, the analogous system based on a BixSn1-xSe constituent was prepared to attempt to systematically affect carrier concentration. It was found that while carrier concentration can be controlled, the evolving structure affects the doping efficiency of the Bi atoms and mobility in the structure. Section 3 outlines attempts to form higher order TiSe2-based heterostructures and the important chemical considerations observed during the preparation of these materials. The 3 component systems in the Pb-Sn-Ti-Se system can be formed at low temperature, with SnSe2 rather than SnSe. One solution is to develop a composite system that couples a nitrogen vacancy (NV) center in diamond to a whispering gallery mode supported by a fused silica microsphere. Such a system is motivated by the optical and electron-spin properties of the NV center. The NV center is the leading spin-qubit and exhibits atomic like linewidths at cryogenic temperatures and has spin coherence times greater than milliseconds at room temperature. These long coherence times, coupled with nanosecond scale spin readout and manipulation times, allow for millions of quantum operations to be processed. Silica whispering gallery resonators are the only class of microresonators with quality factor high enough to reach the strong coupling regime, which is necessary for some quantum information processing applications. Integrating these two components into a system that could position a diamond nanopillar near the surface of a deformed-double stemmed microsphere system, with nanometer precision, at 10 K was a major achievement of this research. Cavity resonances in deformed microspheres can be excited with a free-space coupling technique which simplifies their integration into cryogenic environments. In these intentionally deformed resonators, an enhanced evanescent field decay length was observed at specific locations along the ray orbit. The double-stem arrangement enables the cavity resonance to be tuned over 450 GHz, with sub-10 MHz resolution, at 10 K. These two features, the enhanced decay length and broad range tuning with high resolution, are indispensible tools for cavity-QED studies with silica microspheres. Diamond nanopillars were fabricated from single crystal diamond with diameters as small as 140 nm in order to maintain a high quality factor. Studies were conducted on NV centers in nanopillars and bulk diamond to determine their suitability for cavity-QED applications. The system used for this work was a silica microsphere. In a silica microsphere, the optical cavity is formed by light that is confined by total internal reflection while circulating around the equator of the sphere. The mechanical oscillator is the mechanical breathing motion of the sphere itself. The optical cavity and mechanical oscillator are coupled by radiation pressure and by the mechanical oscillator physically changing the length of the optical cavity. The optomechanical analog to electromagnetically induced transparency (EIT), known as optomechanically induced transparency (OMIT), has previously been studied in its steady state. One topic of this dissertation is an experimental study of OMIT in the time domain. The results of these experimental demonstrations continue comparisons between EIT and OMIT, while also building a foundation for optomechanical light storage. In OMIT, an off-resonance control laser controls the interaction between on-resonance light and the mechanical oscillator. Optomechanical light storage makes use of this arrangement to store an optical signal as a mechanical excitation, which is then retrieved at a later time as an optical signal. This is done by using two temporally separated off-resonance control laser pulses. This technique is extremely flexible in frequency and displays a storage lifetime on the order of microseconds. Use of optomechanical systems for quantum mechanical applications is hindered by the thermal background noise of the mechanical oscillator. Addressing this issue by first cooling the mechanical oscillator is costly and fraught with difficulties. The final topic presented in this dissertation deals with this issue through the use of an optomechanical dark mode. Two optical modes can interact with the same mechanical mode. The dark mode is a state that couples the two optical modes but is decoupled from the mechanical oscillator. While our specific optomechanical system is limited by its somewhat modest optomechanical cooperativity, this conversion process can, in principle, preserve the quantum state of the signal, even at room temperature, opening the possibility for this technique to be applied in quantum information processing. I isotopically evaluate the evolution of magmas from their initial formation, to eruption, and then to their alteration during the diffusion of environmental waters into volcanic glass. I focus first on the formation and evolution of large, caldera-forming eruptions from both Gorely volcano in Kamchatka, Russia and 30–40 Ma caldera forming eruptions through Oregon in the United States of America. I utilize oxygen (δ18O), hafnium (εHf), strontium (87Sr/86Sr), and neodymium (143Nd/144Nd) isotopes to document the creation of caldera-forming eruptions at these eruptive centers through the melting of surrounding crust. I also use U-Pb and 40Ar/39Ar to document the timescales of the formation of these large-volume silicic eruptions. Following eruption, the volcanic glass in tephra and ash can slowly take in environmental water. It is thought that the hydrogen isotopic ratio (δD) of these waters can be used to determine paleoenvironments from the time that the volcanic glass was deposited. The latter portion of my dissertation focuses on the use of hydrogen isotopes of environmentally hydrated volcanic glass to determine paleoenvironments, and the calibration of the TCEA to analyze oxygen isotopes of hydrated volcanic glass. I first focus on the rate of diffusion of water at ambient temperature to better understand the time frame necessary to hydrate volcanic glass for use as a paleoenvironmental indicator. I also document the hydrogen isotopic ratios that result from the diffusion of water into volcanic glass, which is documented as a decrease in δD with an increase in secondary hydration in all regions worldwide except equatorial. Finally, I focus on the earliest stages of diffusion of water into volcanic glass by analyzing tephra deposits that were collected within days of the 1980 eruptions of Mount St. Helens as well as tephra deposits recently collected in 2015 to identify changes in water concentration and hydrogen isotopic ratios over an ~35 year period. Mantle convection is investigated beneath the Galápagos Archipelago and then beneath the Juan de Fuca (JdF) plate, with a focus on the JdF and Gorda Ridges before turning to the off-axis asthenosphere. In the Galápagos Archipelago, S-to-p receiver functions reveal a discontinuity in seismic velocity that is attributed to the dehydration of the upper mantle. The depth at which dehydration occurs is shown to be consistent with prior constraints on mantle temperature. A comparison between results from receiver functions, seismic tomography and petrology shows that mantle upwelling and melt generation occur shallower than the depth of the discontinuity, despite the expectation of high viscosities in the dehydrated layer. Beneath the JdF and Gorda Ridge, low Vs anomalies are too large to be explained by the cooling of the lithosphere and are attributed to partial melt. The asymmetry, large Vs gradients, and sinuosity of the anomalies beneath the JdF Ridge are consistent with models of buoyancy-driven upwelling. However, deformation zone processes appear to dominate mantle flow over seafloor spreading beneath the Explorer and Gorda diffuse plate boundaries. Finally, S-to-p receiver functions reveal a seismic discontinuity beneath the JdF plate that can only be attributed to seismic anisotropy. Synthesis of the receiver function results with prior SKS splitting results requires heterogeneous anisotropy between the crust and the discontinuity. Models of anisotropy feature increasing anisotropy before the decrease at the discontinuity, but well below the base of the lithosphere, and a clockwise rotation of the fast direction with increasing depth. In these results and even in the SKS splitting results, additional driving mechanisms for mantle flow such as density or pressure anomalies are required. While the assembly of the mitotic spindle in C. elegans has been studied in some detail, how the poles assemble in the absence of centrosomes remains poorly understood. In an ongoing screen for temperature-sensitive (TS), embryonic-lethal mutants, we have identified TS mutations in multiple genes required for oocyte meiotic spindle pole assembly. We have so far identified mutations in four genes: or1178ts in mei-1, which encodes the catalytic domain of the microtubule severing complex katanin; or447ts in klp-18, which encodes a kinesin 12 family member; or645ts in aspm-1, which encodes a microtubule scaffolding protein; and or1092ts and or1292ts in klp-7, which encode a kinesin 13/MCAK family member. By using live cell imaging of oocytes from transgenic strains expressing GFP and mCherry fusion to proteins associated with the spindle, we have found and confirmed other findings that klp-18 promotes spindle bipolarity and that MEI-1 promotes pole assembly both by severing microtubules and by recruiting ASPM-1. More recently, we have found that klp-7 is required for maintaining bipolarity in the meiotic spindle by preventing the number of poles that can form. In klp-7(-) mutants, we observed in addition to extraneous poles an excess accumulation of microtubules during Meiosis I. Futhermore, reducing klp-7 function can restore bipolarity in a klp-18(-) monopolar spindle mutant background. We also observed that disruption of the kinetochore factor KNL-1 in klp-7(-) mutants exacerbates the extra spindle pole phenotype. We suggest that in oocyte meiosis, klp-7 is required to limit microtubule accumulation and pole assembly and that it may carry out these functions in a kinetochore-dependent manner. Using a thin-film precursor prepared by sequentially depositing elemental titanium and selenium by physical vapor deposition, the binary compound is readily formed by short-duration annealing at 350°C. The structural and electrical properties of this compound are investigated and suggest that the film is markedly different from the bulk crystalline material. The synthesis is then expanded to produce the first TiSe2 turbostratically disordered misfit layer compound, (PbSe)1.16TiSe2. With the synthesis technique established, the second compound in this family, (PbSe)1.18(TiSe2)2, was synthesized and compared to the misfit layer compound synthesized by bulk methods. The rotationally disordered compound demonstrated an increased Seebeck coefficient and reduced resistivity at room temperature, sparking interest in this family of compounds as potential thermoelectric materials. Designed synthesis of several targeted compounds is reported with interest in evaluating thermoelectric potential of these rotationally disordered compounds. The first nine members of this family were synthesized with m and n values 1 - 3 and evaluated for their thermoelectric properties. Results from this study indicated compounds with larger n values could be of further interest and many more of these compounds are synthesized and reported. Designed synthesis using the MER synthesis method is then pushed further to synthesize the first telluride misfit layer compounds. Four new compounds of the [(PbTe)1.17]m(TiTe2)n family are reported, and their structures were examined by a variety of X-ray diffraction techniques. The robustness of the MER method is revealed by the synthesis of these layered intergrowths which would not be possible using other synthesis methods available.
