The primary research objective was to understand the geochemical interactions of simulated sequestration brines and wellbore cement. There are four key focuses of this study with regards to cement interaction: 1) Simulated brines saturated with Supercritical CO2 and CO2-O2; 2) Adjacent formation host rock under sequestration conditions; 3) Geochemical modeling using CHIM-XPT based on the experimental studies of points 1) and 2) above; 4) Geomechanical strength tests of CO2 exposed cements. First, the interaction of cement with supercritical CO2, CO2-saturated brines, and CO2-O2 mixtures at 50 °C, 85 °C, and 28.9 MPa was evaluated. Cement exposed to a lower salinity brine (1 M) had a larger alteration depth (1.25 mm) compared to cement exposed to a higher salinity brine (2.2 M) (0.836 mm). CO2-O2 exposed cements displayed alteration depths of 1.6 mm at 85 °C and 0.7 mm at 50 °C for 1 M. The cement exposed to the 2.17 M solution had an alteration extending 1.685 mm. The second focus studied the influence of adjacent formations and observed mineral trapping in the Grande Ronde Basalt-cement. Studies of Mt. Simon Sandstone-cement interaction provided evidence for high CO2 storage capacity. The third focus is geochemical modeling using CHIM-XPT. Pure CO2 saturated brines yielded a pH of 3.0 at 1 M and 3.85 at 2.17 M. The CO2-O2 gas run calculated a pH of 3.96 at 1 M and 3.85 at 2.17 M. The Pasco solution after exposure had a pH of 6.97 and an estimated dissolved 161 g of CO2 (g). The pH of the Illinois Basin brine and Mt. Simon sandstone [and cement paste] was 5.8 with an estimated 139 g of CO2 consumed. Lastly, geomechanical tests on CO2 exposed cements indicated that addition of oxygen or changes in temperature play a key role in compressive and tensile strength, decreasing strength by 20-35%. This research highlighted the importance of representing the in-situ wellbore environment when conducting laboratory experiments. Paleocoastal sites on California`s Northern Channel Islands (NCI), dating between ~13,000 and 8000 years ago, may support this coastal migration theory. Until recently, however, we knew little about Paleocoastal technologies, settlement, and lifeways on the islands. Combining traditional archaeological approaches with experimental and archaeometric techniques, I examine Paleocoastal settlement and resource use on San Miguel and Santa Rosa islands. Recently discovered Paleocoastal sites have produced sophisticated chipped stone technologies, with bifacially-flaked points and crescents of extraordinary craftsmanship. Exploring lithic raw material procurement strategies, I demonstrate a Paleocoastal preference for island cherts from sources centered on western Santarosae. Using experimental and archaeometric techniques, I show that Paleocoastal peoples systematically employed heat-treatment to manufacture finely crafted bifaces from island cherts. Using stable oxygen isotope (δ18O) analyses of marine shells from Paleocoastal sites, I examine paleo-sea surface temperatures, seasonality of shellfish collecting, and human sedentism. Evaluating whether such occupations were seasonal or year-round, I tested different sampling strategies for California mussel shells, showing that a method used by many California archaeologists provides erroneous seasonality interpretations for ~35 percent of sampled shells. Using a more intensive sampling strategy, I demonstrate that some Paleocoastal sites were used seasonally, but three substantial middens dating to 8200, 9000, and 10,000 cal BP produced evidence for shell harvesting during all four seasons. This suggests that the NCI were occupied more or less permanently and year-round by at least 10,000 years ago. My research suggests that Paleocoastal peoples had a strong commitment to maritime and island lifeways starting at least 12,000 years ago. From that time until ~8000 years ago, Paleocoastal peoples relied primarily on island resources despite their close proximity to the mainland. The presence of a relatively large, permanent, and distinctive Paleocoastal population on the NCI may also support the coastal migration theory and an even deeper antiquity of human settlement and sedentism on the NCI. How the nervous system does this is a major focus of systems-level neuroscience. This dissertation investigates the neural basis of the sensorimotor transformation underlying a spatial orientation strategy in the nematode Caenorhabditis elegans. Motile organisms rely on spatial orientation strategies to navigate to environments that are conducive to organismal fitness and comfort, e.g. environments with the correct temperature, light level, or access to food and mates. As such, spatial orientation strategies as a class represent a key behavior common to most forms of life on earth. To explore the behavioral mechanism used by C. elegans for spatial orientation, we designed and manufactured a microfluidic device that breaks the feedback loop between self-motion and environmental change by partially restraining the animal. The device takes advantage of laminar flow at small scale to provide distinct environments across the dorsoventral undulation that constitutes locomotion in this animal without using a physical barrier. This device allowed us to conclude that worms use the change in chemical concentration sensed between lateral extremes of the locomotion cycle to direct forward locomotion toward a favorable stimulus, an orientation strategy termed klinotaxis. We then investigated the neuronal basis of this behavior using laser ablation, calcium imaging, and optogenetic stimulation. We found a minimal neuronal network for klinotaxis to sodium chloride including the ASE, AIY, AIZ, and SMB neuron classes that displays left/right asymmetry across the sensory neuron, interneuron, and motor neuron levels. We extended these results by ablating other neurons that have been implicated in klinotaxis in other studies. Finally, we imaged the ASE neurons during klinotaxis in microfluidic device and found that these neurons are active on the timescale of individual head swings. Additionally, we found anecdotal evidence that photostimulation of ASE neurons expressing the light sensitive ion channel Channel Rhodopsin (CHR2) is sufficient to stimulate klinotaxis behavior.
