Year: 2013 Pine Creek Conservation Area (PCCA), just northeast of the John Day River in Wheeler County, Oregon, was acquired in 1999-2001 by the Confederate Tribes of Warm Springs with support from the Bonneville Power Administration (BPA), to mitigate for wildlife losses created by the large, hydropower Columbia River Dams, particularly the Bonneville, Dalles and John Day Dams. Many thousands of acres of grassland, shrub steppe and riparian habitats were lost due to inundation, and the objectives of the acquisition included restoration of similar habitats. As part of an interagency agreement created in 2002, the Oregon Natural Heritage Information Center established a baseline monitoring program in 2002. This effort included the establishment of permanent monitoring plots to allow for a detailed assessment of vegetation change in the plant communities occurring at the site. It also included the development of an existing vegetation map, hopefully to allow for an analysis of overall vegetation change across the conservation area. The map showed the distribution of western juniper, native grasslands, big sagebrush, and weed-dominated areas at the site. In the eight years since the original map was made, a series of management actions, including juniper clearing, prescribed fires, and riparian restoration activities have significantly changed the vegetation at PCCA. In the spring and summer of 2010, the Oregon Natural Heritage Information Center, now the Oregon Biodiversity Information Center at Portland State University, visited the area to assist the land manager in developing a strategy for meeting the information needs of the Tribes and BPA in evaluating the success of the first decade of restoration. This report details that effort, which incorporated a combination of field inspection, photo-interpretation, and remote sensing-based mapping to assess change since the establishment of the conservation area, to lay a new baseline against which to measure future change, and most importantly to provide detailed information useful for land management decision making in the continuing restoration efforts. Thus, it is important to consider what constitutes an adequate sample not only for all taxa as a whole but also for each taxon individually as well as how to strategically schedule collection over time. There are five basic sampling questions: from which species to collect, from how many and which populations, from how many and which individuals, how many and what kind of propagules to collect, and, finally, at what point is the desired sample size too great for a population to bear in one year? There is no single correct sampling strategy or protocol. Each situation must be evaluated in the context of the particular purposes, goals, and uses for which the collection is being made as well as the nature of the sampling universe, the manner in which the samples will be stored and maintained, the period of time they will be stored, and whether the wild-collected samples are to be used directly or whether their numbers will be increased by agricultural growouts. Purposes include providing a long-term “insurance policy” against extinction in the wild and, in the near to medium term, supplying native plant material for small- to large-scale restoration purposes. The term “sampling universe” refers to the target taxa of interest: it could be the entire seed plant flora of Taiwan or particular subsets, such as endemics, higher-elevation plants, ecologically important taxa, rare plants, and so on. Plants with orthodox seeds are much easier to work with and are more economically stored as dried, frozen seeds in a seed bank than either plants with recalcitrant seeds or taxa maintained as growing plants. Securing both species and ecosystem services might be accomplished with common solutions. Yet it is unknown whether these two major conservation objectives coincide broadly enough worldwide to enable global strategies for both goals to gain synergy. In this article, we assess the concordance between these two objectives, explore how the concordance varies across different regions, and examine the global potential for safeguarding biodiversity and ecosystem services simultaneously. We find that published global priority maps for biodiversity conservation harbor a disproportionate share of estimated terrestrial ecosystem service value (ESV). Overlap of biodiversity priorities and ESV varies among regions, and in areas that have high biodiversity priority but low ESV, specialized conservation approaches are necessary. Overall, however, our findings suggest opportunities for safeguarding both biodiversity and ecosystem services. Sensitivity analyses indicate that results are robust to known limitations of available ESV data. Capitalizing on these opportunities will require the identification of synergies at fine scales, and the development of economic and policy tools to exploit them. This study's lithic debitage analysis, and investigation of hunter-gatherer technological organization through time, contributes to the knowledge base and understanding of how hunter-gatherers used subalpine environments in MORA. The debitage sample is from archaeological excavations between 2005 and 2007 at a Buck Lake Site (45PI438) activity area in the subalpine environmental zone. Two cultural components were examined: the pre-Mount St. Helens Yn tephra component (before 3500 RCYBP) is thought to represent a forager-like mobility strategy and the post-Mount St. Helens Yn tephra component (after 3500 RCYBP) is thought to represent a collector-like strategy. Expectations theoretically grounded in hunter-gatherer mobility, tool design, raw material procurement, site function, and tool function were developed and tested. Results suggest that hunter-gatherers at Buck Lake relied on and maintained small, lightweight, transported bifaces made of nonlocal raw material regardless of expected changes in mobility strategy through time. For both foragers and collectors at Buck Lake, similar lithic raw material availability, terrain, and seasonality constraints and a common resource acquisition goal and overlapping site function resulted in similar hunter-gatherer technological organization strategies. Slight differences between the cultural components include: the use of more local igneous raw material in the forager-like component, the use of a more expedient technology in the collector-like cultural component, and smaller size debitage in the forager-like component. The use of expedient bipolar technology in both cultural components is possible, but only partially supported. Evidence of bipolar technology would suggest that hunter-gatherers were conserving nonlocal CCS by using the bipolar technique on exhausted transported tools or cached cores to produce expedient flakes used for small-game hunting and processing. Further research for the Buck Lake site should include: the sourcing of raw material; conducting experimental lithic reduction on toolstone found at Buck Lake to produce comparative debitage specimens; and increasing the lithic analysis sample size to include debitage recovered from 2008-2009 excavations and other artifact types.   Citation × Citation Citation Abstract Copy Title: "As Long as the Mighty Columbia River Flows": The Leadership and Legacy of Wilson Charley, a Yakama Indian Fisherman Author: Hedberg, David-Paul Brewster Year: 2017 On March 10, 1957, the United States Army Corps of Engineers completed The Dalles Dam and inundated Celilo Falls, the oldest continuously inhabited site in North America and a cultural and economic hub for Indigenous people. In the negotiation of treaties between the United States, nearly one hundred years earlier, Indigenous leaders reserved access to Columbia River fishing sites as they ceded territory and retained smaller reservations. In the years before the dam's completion, leaders, many of who were the descendants of earlier treaty signatories, attempted to stop the dam and protect both fishing sites from the encroachment of state and federal regulations and archaeological sites from destruction. This study traces the work of Wilson Charley, a Native fisherman, a member of the Yakama Nation's Tribal Council, and great-grandson of one of the 1855 treaty signatories. More broadly, this study places Indigenous actors on a twentieth-century Columbia River while demonstrating that they played active roles in the protest and management of areas affected by The Dalles Dam. Using previously untapped archival sources--a substantial cache of letters--my analysis illustrates that Charley articulated multiple strategies to fight The Dalles Dam and regulations to curtail Native's treaty fishing rights. Aiming to protect the 1855 treaty and stop The Dalles Dam, Charley created Native-centered regulatory agencies. He worked directly with politicians and supported political candidates, like Richard Neuberger, that favored Native concerns. He attempted to build partnerships with archaeologists and landscape preservationists concerned about losing the area's rich cultural sites. Even after the dam's completion, he conceptualized multiple tribal economic development plans that would allow for Natives' cultural and economic survival. Given the national rise of technological optimism and the willingness for the federal government to terminate its relationship with federally recognized tribes, Charley realized that taking the 1855 treaty to court was too risky for the political climate of the 1950s. Instead, he framed his strategies in the language of twentieth-century conservation, specifically to garner support from a national audience of non-natives interested in protecting landscapes from industrial development. While many of these non-native partners ultimately failed him, his strategies are noteworthy for three reasons. First, he cast the fight to uphold Native treaty rights in terms that were relevant to non-natives, demonstrating his complex understanding of the times in which he lived. Second, his strategies continued an ongoing struggle for Natives to fish at their treaty-protected sites, thereby documenting an overlooked period between the fishing rights cases of the turn of the twentieth century and the 1960s and 1970s. Charley left a lasting legacy that scholars have not recognized because many of his visionary ideas came to fruition decades later. Finally, my analysis of Charley's letters also documents personal details that afford readers the unique perspective of one Indigenous person navigated through a tumultuous period in the Pacific Northwest and Native American history. The type IB GTP cyclohydrolase (GCYH-IB) is a prokaryotic-specific enzyme found in a number of pathogens. GCYH-IB is structurally distinct from the canonical type IA GTP cyclohydrolase involved in biopterin biosynthesis in humans and animals, and thus is of interest as a potential antibacterial drug target. We report kinetic and inhibition data of Neisseria gonorrhoeae GCYH-IB, and two high-resolution crystal structures of the enzyme; one in complex with the reaction intermediate analog and competitive inhibitor 8-oxo-GTP, and one with a TRIS molecule bound in the active site and mimicking another reaction intermediate. Comparison with the type IA enzyme bound to 8-oxo-GTP reveals an inverted mode of binding of the inhibitor ribosyl moiety and, together with site-directed mutagenesis data, shows that the two enzymes utilize different strategies for catalysis. Notably, the inhibitor interacts with a conserved active site Cys149, and this residue is S-nitrosylated in the structures. This is the first structural characterization of a biologically S-nitrosylated bacterial protein. Mutagenesis and biochemical analyses demonstrate that Cys149 is essential for the cyclohydrolase reaction, and S-nitrosylation maintains enzyme activity, suggesting a potential role of the S-nitrosothiol in catalysis.
