The results indicate that water in the fish tanks must be slightly acid (pH 6), warm (22°C), nonmoving, and that the fish must be wounded in order for fungal infection to occur. Cool water (18°C) and moving water, caused by aeration and filtration, are detrimental to fungal colony formation so that no infection occurs. Both S. ferax and S. parasitica infected both species of fish, on an average of three days after innoculation. The S. ferax infected a higher percentage of both fishes than did the S. parasitica. Year: 1984 Goldfish (Carassius auratus) of 15-28g were maintained at 23 C and subsequently were intraperitoneally injected with saline (controls) and varying doses of Triiodothyronine (T3). Following the injection, the fish were placed in a thermal gradient and temperature selection and activity monitored for 30 min. There was no- significant difference between the responses of the experimental fish to the 2 different doses of T3. However, there was a significant difference in response between the experimental and control groups (P < 0.001). The decreased temperature selection occurred early in the test runs, while the increased activity began later. Larger goldfish (175-205g) received 0.5 μl intracerebral injections of 1.5-3.0 ng T3 in the anterior brainstem via an indwelling cannula injection system. These fish responded to the injections by selecting cooler water. The magnitude of the response was related to the distance of the injection site from the preoptic/anterior hypothalamic region. I conclude that increased plasma levels of T3 lead to the selection of cooler water. This effect appeared to be mediated via the anterior brainstem. Injected T3 also produced hyperactivity. This latter effect developed more slowly and appeared to be more diffusely mediated.   Citation × Citation Citation Abstract Copy Title: Effect of ethanol on thermoregulation in the goldfish, Carassius auratus Author: O'Connor, Candace Sharon Year: 1986 In an attempt to elucidate the mechanism by which ethanol affects vertebrate thermoregulation, the effect of ethanol on temperature selection was studied in the goldfish, Carassius auratus. Ethanol was administered to 10 to 15 g fish by mixing it in the water of a temperature gradient. The dose response curve was very steep between 0.5% (v/v) ethanol (no response) and 0.7% (significant lowering of selected temperature in treated fish). Fish were exposed to concentrations of ethanol as high as 1.7%, at which concentration most experimental fish lost their ability to swim upright in the water. At concentrations higher than 0.7%, the magnitude of the effect did not increase with increasing concentration of ethanol; treated animals continued to select temperatures about 2 C below temperatures selected by controls. Experiments alternating exposure to 1.0% ethanol and water showed that the rate of onset and disappearance of the ethanol effect was rapid (within 10 min). Other experiments exposing fish to 1.0% ethanol for up to 3 hr showed that the effect remained stable for this period of time. The thermoregulatory responses of fish are behavioral, and therefore relatively easy to observe and quantify. Ethanol produces a prompt, stable and reproducible depression of selected temperature in the goldfish. Because the temperature at which fish regulate is controlled by a central nervous system set point and not altered by effects on peripheral effector systems, it appears that ethanol may cause hypothermia in goldfish by directly acting to lower the set point. The possibility of attempting to use human strains of herpesviruses in order to transform normal fish tissue culture preparations was considered. Three different fish cell cultures were tested: CHSE-214 cell lines were derived from embryonic chinook salmon (Oncorhynohus tshawytscha), and STE-137 cell lines were obtained from embryonic steelhead trout (Salmo gairdneri). Cells isolated from immature ovaries of rainbow trout (Salmo gairdneri) were propagated by the investigator in these studies. Both HSV 1 and HSV 2 demonstrated cytopathic effects on the 3 different cell types described above when these cells were observed during the exponential growth phase. If virus was added before cells were dividing actively, or when the cells were maintained in a stationary phase of growth, no effect was observed. Several methods were employed in an attempt to trans- form fish cells grown in tissue culture. Different quantities of live virus, preparations of ultraviolet light in- activated virus and purified viral DNA were used in these studies but malignant conversion by known viral components was not achieved. In these experiments, the 2B strain of cells obtained originally from immature ovaries of rainbow trout appeared to undergo spontaneous transformation thereby becoming established as a permanent cell line. To date tissue cultures designated 2B have undergone 25 transfers in this laboratory. It reviews the most recent research on acidic, Antarctic, cave, desert, hypersaline, hypoxic, temporary, and fast-flowing habitats, as well as naturally and anthropogenically toxic waters, while pointing out generalities that are evident across different study systems. Knowledge of the different adaptations that allow fish to cope with stressful environmental conditions furthers our understanding of basic physiological, ecological, and evolutionary principles. In several cases, evidence is provided for how the adaptation to extreme environments promotes the emergence of new species. Furthermore, a link is made to conservation biology, and how human activities have exacerbated existing extreme environments and created new ones. The book concludes with a discussion of major open questions in our understanding of the ecology and evolution of life in extreme environments. Zebrafish (Danio rerio) are used a model organism because of their highly fecundity, external fertilization, and robust nature, making them highly adaptable to environmental and genetic variation. In an effort to limit data variation that lies outside of topic interest, phenotypic measures of variation must be performed, understood, and taken into consideration for future studies. A common measurement of phenotypic variation in fish is in the maximum (Ucrit) swimming speeds. Inter and intra-strain variation in zebrafish Ucrit swimming speeds will be observed in a swim tunnel. Baseline values will be recorded from ten fish from each of two families in each of three strains: WIK, Tubigan and AB. Environmental variation in zebrafish raised (beginning at one month of age) in tanks with various volumes and densities of fish will also be recorded from each strain.
