There is a looming concern that non-farm related uses and dwellings, or non-farm development, are conflicting with agricultural preservation strategies. Specifically, non-farm developments can potentially undermine the critical mass of farmland needed to keep the agricultural economy sustainable, but until now, studies have lacked spatially precise data to systematically track these phenomena. This thesis offers methodological contributions towards analyzing these operations and presents a broad account of what has been occurring in the region. Using permit approval data from the Department of Land Conservation and Development (DLCD) and 2015 county tax lot shapefiles, I geocoded the locations of these uses and dwellings. I used location quotient and spatial autocorrelation coefficients to identify non-farm hotspots in the region and summarized different typologies that have developed. The findings reveal that viticulture operations have amassed near Dundee and Newberg in Yamhill County, while commercial activities and home occupations have clustered near the Salem-Keizer UGB. Concurrently, dwellings have clustered near the Yamhill-Polk County border. Finally, I offer suggestions to improve Oregon's agricultural land use policy and data management process, as well as advocate for more intensive research in the future to generate narratives for our results. The type IB GTP cyclohydrolase (GCYH-IB) is a prokaryotic-specific enzyme found in a number of pathogens. GCYH-IB is structurally distinct from the canonical type IA GTP cyclohydrolase involved in biopterin biosynthesis in humans and animals, and thus is of interest as a potential antibacterial drug target. We report kinetic and inhibition data of Neisseria gonorrhoeae GCYH-IB, and two high-resolution crystal structures of the enzyme; one in complex with the reaction intermediate analog and competitive inhibitor 8-oxo-GTP, and one with a TRIS molecule bound in the active site and mimicking another reaction intermediate. Comparison with the type IA enzyme bound to 8-oxo-GTP reveals an inverted mode of binding of the inhibitor ribosyl moiety and, together with site-directed mutagenesis data, shows that the two enzymes utilize different strategies for catalysis. Notably, the inhibitor interacts with a conserved active site Cys149, and this residue is S-nitrosylated in the structures. This is the first structural characterization of a biologically S-nitrosylated bacterial protein. Mutagenesis and biochemical analyses demonstrate that Cys149 is essential for the cyclohydrolase reaction, and S-nitrosylation maintains enzyme activity, suggesting a potential role of the S-nitrosothiol in catalysis. LiDAR technology’s use on UAS (Unmanned Aerial System) is a relatively new concept in the rapidly advancing remote sensing & 3D imaging industry. As such, profitability data is either not released by similar business organizations, or there is simply not enough historical data to accurately predict revenue streams with either business model. To assess the feasibility of business models, market research and literature review were performed by our research team. Economic analysis methods were used to determine the most advantageous business plan strategy for the LiDAR business opportunities. Since the success of either business model will rely heavily on the company’s sales ability. Since the focus of this report is on economic analysis to make the best business decision, we have made our valuation calculations based conservative sales forecasts. The startup business plan identifies and critiques two business models identified by the founder entrepreneurs: 1. A service oriented model in which data is collected, processed, and delivered by an operations team to the customer. 2. A product oriented model in which LiDAR UAS is constructed, tested, and sold as a unit that includes training as an entire product for the customer’s own use. Based on these two models, three decision options were evaluated and compared with each other: 1. Invest in the Service only model 2. Invest in the Product only model 3. Do not invest, utilize the stock market for investment gains A 5-year cash flow analysis was conducted to give the founders a recommended strategy to optimize their investment. Through conducting a benefit cost analysis and calculating the internal rate of return of each opportunity, it was determined that both business models may be profitable and worthy investments. The expected IRR of service model and product model are 56% and 136% respectively. They are both much higher than the stock market option, which has an estimated MARR of 8% on average. Much of the risk in this analysis was the uncertainty in the sales revenue projections. The team endeavored to determine the sensitivity of the data by performing sensitivity analysis on these sales projections. Expected values were calculated taking into consideration of the most likely, optimistic and pessimistic cases. The sensitivity analysis result shows both business models are profitable under all conditions. The economic analysis has shown that while both business models are likely to be profitable, the Product Business Model has the potential for a higher return on investment. Therefore the research team recommends that the investors proceed with the Product Business Model outlined in the report. However, despite substantial advances in our understanding of work engagement at the between-person level, scholars have argued for increased investigation into what drives engagement on a daily level for individual employees. In the current study, a within-person, day-level design was employed to examine the relationships between nonwork mastery experiences, job crafting behaviors, and daily work engagement. Drawing on Conservation of Resources (Hobfoll, 1989) theory, nonwork mastery experiences and job crafting were operationalized as employee-driven, resource-building strategies that assist employees in generating important psychological and job resources that can be drawn upon in order to maintain high levels of work engagement during the day. Moreover, a reciprocal relationship between work engagement during the day and nonwork mastery experiences the same evening was tested. Employees from a U.S. technology firm provided responses in the morning, at lunchtime, and after work each day for five working days. Multilevel structural equation modeling was used to test the hypotheses in the current study. Findings revealed no support for the hypothesized model at the within-person level of analysis; however, ancillary analyses suggested support for an indirect relationship between job crafting and work engagement via increased positive affect. Moreover, nearly all the proposed relationships emerged at the between-person level of analysis providing some insight into the effects of resource building strategies and work engagement across participants. Finally, seeking structural resources was identified as a person-level factor that explained variance in employees' initial levels of work engagement at the start of the week, as well as the trajectory of engagement over the course of the week. The current findings contribute to our understanding of bottom-up, employee-driven behaviors that help to sustain engagement over time. Suggestions for future research and implications for practice are discussed. Understanding the biology and environmental tolerances of dreissenid mussels, and effectiveness of various man­agement strategies, is key to prevention. On November 4-5, 2015, the Aquatic Bioinvasion Research and Policy Institute and the Center for Lakes and Reservoirs at Portland State University, the US Geological Survey, and the Pacific States Marine Fisheries Commission, convened a Dreissenid Mussel Research Priorities Workshop funded by the Great Northern Landscape Conservation Cooperative. The purpose of the workshop was to review dreissenid research priorities in the 2010 Quagga-Zebra Mussel Action Plan for Western U.S. Waters, reassess those priorities, incorporate new information and emerging trends, and develop priorities to strategically focus research efforts on zebra and quagga mussels in the Pacific Northwest and ensure that future research is focused on the highest priorities. It is important to note that there is some repetition among dreissenid research priority categories (e.g., prevention, detection, control, monitoring, and biology). Workshop participants with research experience in dreissenid mussel biology and management were identified by a literature review. State and federal agency managers were also invited to the workshop to ensure relevancy and practicality of the work­shop outcomes. A total of 28 experts (see sidebar) in mussel biology, ecology, and management attended the workshop. Methods We used a novel integration of a forest landscape succession and disturbance model (LAN DIS-II) with a fine-scale climatic water deficit approach to simulate dynamics of aspen and associated conifer and shrub species over the next 150 years under various climate-fire scenarios. Results Simulations suggest that many aspen stands could persist without fire for centuries under current climate conditions. However, a simulated 2–5ºC increase in temperature caused a substantial reduction of aspen coverage at lower elevations and a modest increase at upper elevations, leading to an overall reduction of aspen range at the landscape level. Increasing fire activity may favor aspen increase at its upper elevation limits adjacent to coniferous forest, but may also favor reduction of aspen at lower elevation limits adjacent to xeric shrubland. Conclusions Our study highlights the importance of incorporating fine-scale terrain effects on climatic water deficit and ecohydrology when modeling species distribution response to climate change. This modeling study suggests that climate mitigation and adaptation strategies that use fire would benefit from consideration of spatial context at landscape scales. Thus, it is important to consider what constitutes an adequate sample not only for all taxa as a whole but also for each taxon individually as well as how to strategically schedule collection over time. There are five basic sampling questions: from which species to collect, from how many and which populations, from how many and which individuals, how many and what kind of propagules to collect, and, finally, at what point is the desired sample size too great for a population to bear in one year? There is no single correct sampling strategy or protocol. Each situation must be evaluated in the context of the particular purposes, goals, and uses for which the collection is being made as well as the nature of the sampling universe, the manner in which the samples will be stored and maintained, the period of time they will be stored, and whether the wild-collected samples are to be used directly or whether their numbers will be increased by agricultural growouts. Purposes include providing a long-term “insurance policy” against extinction in the wild and, in the near to medium term, supplying native plant material for small- to large-scale restoration purposes. In the negotiation of treaties between the United States, nearly one hundred years earlier, Indigenous leaders reserved access to Columbia River fishing sites as they ceded territory and retained smaller reservations. In the years before the dam's completion, leaders, many of who were the descendants of earlier treaty signatories, attempted to stop the dam and protect both fishing sites from the encroachment of state and federal regulations and archaeological sites from destruction. This study traces the work of Wilson Charley, a Native fisherman, a member of the Yakama Nation's Tribal Council, and great-grandson of one of the 1855 treaty signatories. More broadly, this study places Indigenous actors on a twentieth-century Columbia River while demonstrating that they played active roles in the protest and management of areas affected by The Dalles Dam. Using previously untapped archival sources--a substantial cache of letters--my analysis illustrates that Charley articulated multiple strategies to fight The Dalles Dam and regulations to curtail Native's treaty fishing rights. Aiming to protect the 1855 treaty and stop The Dalles Dam, Charley created Native-centered regulatory agencies. He worked directly with politicians and supported political candidates, like Richard Neuberger, that favored Native concerns. He attempted to build partnerships with archaeologists and landscape preservationists concerned about losing the area's rich cultural sites. Even after the dam's completion, he conceptualized multiple tribal economic development plans that would allow for Natives' cultural and economic survival. Given the national rise of technological optimism and the willingness for the federal government to terminate its relationship with federally recognized tribes, Charley realized that taking the 1855 treaty to court was too risky for the political climate of the 1950s. Instead, he framed his strategies in the language of twentieth-century conservation, specifically to garner support from a national audience of non-natives interested in protecting landscapes from industrial development. While many of these non-native partners ultimately failed him, his strategies are noteworthy for three reasons. First, he cast the fight to uphold Native treaty rights in terms that were relevant to non-natives, demonstrating his complex understanding of the times in which he lived. Second, his strategies continued an ongoing struggle for Natives to fish at their treaty-protected sites, thereby documenting an overlooked period between the fishing rights cases of the turn of the twentieth century and the 1960s and 1970s. Charley left a lasting legacy that scholars have not recognized because many of his visionary ideas came to fruition decades later. By using the water tanks as a thermal storage unit, this project attempts to partially decouple energy consumption from generation to provide peak demand reduction and to better facilitate the integration of variable renewable energy resources. A scoping study was performed to evaluate the potential impact of this project if implemented in single family homes in Portland, Oregon. This study revealed that the system could provide meaningful savings in the cost of electricity to both the customer and utility. Additionally, an optimization algorithm was developed to dictate system operation and to maximize gains to the utility. Evolutionary algorithms were explored in an attempt to increase the effectiveness of the algorithm's search in limited computation time. Ultimately, an evolution strategy was selected as the most suitable based on tests run in winter and spring months. A genetic algorithm was then developed to handle fixed-speed heat pump operation for compatibility with an alpha-system prototype that has been developed by the research team.
