The current design approach for structures constructed with this type of wall form only allows the designer to consider the contribution of the reinforced concrete cores. Previous research has shown that this approach may be conservative. This project experimentally evaluated the lateral structural response of these types of grid ICF walls under increasing amplitude of in-plane cyclic loading. Two different height-to-length (aspect) ratios (approximately 2:1 and 1:1) were investigated, as was the effect of simultaneous gravity load. Furthermore, the reinforced concrete grid was exposed for each aspect ratio in order to examine the contribution of the ICF blocks to the lateral response. Analyses of hysteretic behaviors and failure modes indicated conservatism in the current design approach for estimating lateral strength and ignoring the beneficial contribution of the ICF blocks to overall performance. The presence of the wall forms increased the lateral shear capacity of the walls by an average of 42% (compared to the walls with forms removed), while also increasing the deformation capacity by an average of 102%. Furthermore, by considering an additional gravity load of 10 kips-per-lineal-foot (klf), the shear resistance of the walls increased by 32% (versus walls only subjected to self-weight), on average, and the deformation capacity of the walls increased by an average of 19%. Comparisons of the experimental results to several design equations led to the recommendation of a design equation that was previously accepted for another type of ICF system. In terms of attitudes, two attitudinal measures emerged from these data following a Cronbach's alpha and Confirmatory Factor Analysis (CFA): the pro-environmental attitude and concern for the energy use in the U.S. society. These two attitudes, along with other socio-demographic and external factors (home ownership, weather, price of energy, etc.), were examined to assess whether attitude-behavior relationships persisted over time, were more prominent across certain groups, or were constrained by income or other socio-demographic factors. Three theoretical viewpoints of how attitudes may relate to behavior guided the analysis on how attitudes and contextual factors may inter-relate either directly or through a moderator variable to affect thermostat-setting and CFL installation behavior. Results from these analyses revealed four important patterns. First, a relationship between the pro-environmental attitude and the two behaviors (thermostat-setting and CFL installation behavior) was weak but persistent across time. Second, financial factors such as income moderated the pro-environmental attitude and CFL installation relationship, indicating that the pro-environmental attitude could influence the behavior in those situations where financial resources are sufficient to comfortably allow the consumer to participate. Third, this study documented that most people reported changing thermostat settings to save energy or having one or more CFLs in their homes. This finding suggests that organizations, policy makers, or energy efficiency program administrators may want to assess whether they should pursue these two behaviors further, since they appear to be very common in the U.S. population. Last, this study showed that thermostat-setting and CFL installation behavior have multi-factorial influences; many factors in addition to attitudes were significantly associated with these behaviors, and all these factors together explained no more than 16% of behavioral variance. This suggested that if energy-saving behaviors are a function of many different variables, of which none appear to be the "silver bullet" in explaining the behaviors (as noted in this study), then policy analysis should explore a broader number of causal pathways and entertain a wider range of interventions to influence consumers to save energy. TFB1 is very similar to TFIIB in terms of sequence homology and function, whereas TFB2 is unusual as it is missing highly conserved sequences in its N-terminal domain that are present in TFIIB and TFB1. Despite this, TFB2 is effective in transcription process, albeit with lower efficiency compared to TFB1. Other archaea also contain multiple TFBs, but unlike Pyrococcus furiosus TFB2, these multiple TFBs have higher sequence homology to each other and have similar transcription efficiencies. Photochemical cross-linking experiments have shown that the B-reader of TFB in archaea and TFIIB in eukaryotes is close to transcription start site and is very important in RNAP recruitment to promoter DNA and transcription start site selection. The B-reader loop region is close to transcription start site and interacts with the transcribed strand of promoter DNA while the B-linker strand cross-links with the non-transcribed strand. Some of the amino acids in between the B-reader loop and the B-linker strand region in TFB2 are seen to cross-link both the transcribed and the non-transcribed strand. Thus, despite the absence of strong homology to conserved B-reader and B-linker sequences, TFB2 is likely to interact with DNA in the transcription bubble and facilitate in transcription initiation. Utilities typically size their transformers based on a formula that takes into account broadly what types and how many dwellings will be connected. Most new residential dwellings feature 200 Amp service per household with an anticipated energy demand of under 20,000 kWh per year. Average electrical energy consumption varies from state to state but averages to 11,280 kWh per year. Energy demand is expected to fall into a typical residential load curve that shows increased demand early in the morning, then decreasing during the day and another peak early to late evening. Distribution transformers are sized at the limit of the combined evening peak with the assumption that the transformer has enough thermal mass to absorb short overloads that may occur when concurrent loading situations among multiple dwellings arise. The assumption that concurrent loading is of short duration and the transformer can cool off during the night time has been validated over the years and has become standard practice. This has worked well when dwelling loads follow an averaging scheme and low level of coincidence. With the arrival of electric vehicles (EV's) this assumption has to be reevaluated. The acquisition of an electric vehicle in a household can drive up energy demand by over 4000 kWh per year. Potentially problematic is the increased capacity of battery packs and the resulting proliferation of Level 2 chargers. The additional load of a single Level 2 charger concurring with the combined evening peak load will push even conservatively sized distribution transformers over their nameplate rating for a substantial amount of time. Additionally, unlike common household appliances of similar power requirements such as ovens or water heaters, a Level 2 battery charger will run at peak power consumption for several hours, and the current drawn by the EVs has very high levels of harmonic distortion. The excessive loading and harmonic profile can potentially result in damaging heat build-up resulting in asset degradation. In this thesis I present a device and method that monitors pole mounted distribution transformers for overheating, collect and wirelessly upload data and initiate commands to chargers to change output levels from Level 2 to Level 1 or shut down EV charging altogether until the transformer returns into safe operational range. Year: 2014 Successful captive-breeding and re-introduction programs must have the ability to breed a surplus of genetically suitable animals for release into the wild. Unfortunately, many individuals in captive breeding programs often do not reproduce even when they are apparently healthy and presented with genetically appropriate mates. Mate choice can affect multiple parameters of reproductive sperformance, including mating success, offspring production, survival, and fecundity. We investigated the role of mate preference and personality on the reproductive performance of male and female giant pandas (Ailuropoda melanoleuca) as measured by intromission success and litter production. We conducted these studies on giant pandas at the China Conservation and Research Center for the Giant Panda (Bifengxia, Sichuan, China.) from February-May 2011, 2012, and 2013. In Chapter 2, we compared reproductive performance between individuals mated with preferred and nonpreferred males of the focal female (female preference), the focal male (male preference), and mutual mate choice perspective. There were four types of reproductive pairings in our mutual mate choice analyses: females and males that preferred each other (P-P), females that preferred the male but the male did not prefer the female (P-NP), females that did not prefer the male but the male preferred the female (NP-P), and females and males that did not prefer each other (NP-NP). Pairing giant pandas with a preferred partner increased the probability of intromission success and producing a cub. Of the 25 females mated to a preferred partner, 72% had successful intromission, 64% produced cubs, and 52% reared their own cubs versus 31%, 12%, 12% for females mated to their nonpreferred partner (N = 16). Male giant panda mate preference showed similar results. Of the 24 males that were mated with their preferred females, 75% had successful intromission, 77.8% produced cubs, and 92.9% produced cubs that were maternally reared versus 31%, 60%, 66.7% for males mated with their nonpreferred females (N = 16). There was a statistically significant difference in intromission success and cub production for P-P pairings versus NP-NP pairings. Of the twelve P-P mate dyads 83% had successful intromission, 90% produced cubs, and 88.9% had mothers rear their cubs. Of the four P-NP mate dyads 50% had successful intromission, 50% produced cubs, and 100% had the mother rear their cub. Of the seven NP-P mate dyads 57% had successful intromission, 50% produced cubs, and 100% had the mothers rear their cubs. Of the three NP-NP mate dyads 0% had successful intromission resulting in 0% producing cubs and having mother-rear cubs. In addition, we took into account various life and breeding history factors that may have affected reproductive performance such as birth location, rearing, previous sexual history with the opposite sex and familiarity status directly prior to breeding. Mate pairings that were familiar with each other as measured by successfully producing a cub in the past, had increased intromission success. Females mated to heavier males had more intromissions and produced more cubs than females mated to smaller males. In Chapter 3 and 4, we investigated the effect of personality on reproductive performance measurements. Recently, personality of individual animals has been implicated in the failure or success of captive management and breeding of threatened and endangered species. Measurements of personality included novel object tests (where behaviors of individual giant pandas were scored toward a novel object for one hour) and keeper surveys (where the primary keeper rated giant pandas on 23 adjectives describing personality); both measures significantly correlated with variation in long-term and specific mating dyad reproductive performance. In Chapter 3 we evaluated the effect of personality on long-term reproductive performance. Our results show that for both male and female giant pandas, `fearful' individuals had lower natural mating success and cub production. In addition, `aggressive' female giant pandas had increased natural mating success but decreased cub production. Males did not show these relationships between `aggressive' personality types and reproductive performance. Female giant pandas that were labeled as `aggressive' had higher frequencies of positive mating behaviors but `fearful of conspecifics' females had lower frequencies. Males labeled `fearful of conspecifics' and with high `playful-clever' component scores had a lower frequency of positive mating behaviors. In Chapter 4 we further investigated the effects of personality within a specific mating dyad. Recent studies suggest that mate compatibility within personality types can improve reproductive performance measurements such as mating success, offspring survivorship, and offspring quality. We investigated the effect of personality compatibility on reproductive performance through analyzing mate dyad similarity in personality (similar versus dissimilar), specific personality combinations, and relative personality scores (male scores higher or lower than female scores). The specific personality combinations were: females and males that were both low on the personality component (L_L), females were low but males were high (L_H), females were high but males were low (H_L), and females and males were both high (H_H). Our results suggest that giant pandas appear to be assortatively mating based on aggressive, fearful, and stereotypical-excitable personality traits. Mate dyads that were dissimilar on the stereotypical-excitable component score were more likely to have successful intromission than pairs that were similar. L_H mate dyads on the aggressive and stereotypical-excitable personality traits had increased intromission success and cub production than all other possible personality combinations. L_L on fearfulness had higher intromission success but not higher cub production than all other possible personality combinations. Mate dyads composed of males ranked high on the aggressive component relative to the female resulted in higher reproductive performance and cub production. In captive breeding programs, mates are traditionally selected solely on the basis of genetic parameters to minimize loss of genetic diversity and inbreeding coefficients. Our results suggest that integrating genetic with behavioral measures of preference, breeding history factors, and personality would increase the reproductive output of the giant panda captive-breeding program. We stress the importance of considering husbandry practices to accommodate personality traits and ultimately increase reproductive performance in the captive population while simultaneously maintaining both genetic and behavioral diversity. In Chapter 3 and 4, we investigated the effect of personality on reproductive performance measurements. Recently, personality of individual animals has been implicated in the failure or success of captive management and breeding of threatened and endangered species. Measurements of personality included novel object tests (where behaviors of individual giant pandas were scored toward a novel object for one hour) and keeper surveys (where the primary keeper rated giant pandas on 23 adjectives describing personality); both measures significantly correlated with variation in long-term and specific mating dyad reproductive performance. In Chapter 3 we evaluated the effect of personality on long-term reproductive performance. Our results show that for both male and female giant pandas, `fearful' individuals had lower natural mating success and cub production. In addition, `aggressive' female giant pandas had increased natural mating success but decreased cub production. Males did not show these relationships between `aggressive' personality types and reproductive performance. Female giant pandas that were labeled as `aggressive' had higher frequencies of positive mating behaviors but `fearful of conspecifics' females had lower frequencies. Males labeled `fearful of conspecifics' and with high `playful-clever' component scores had a lower frequency of positive mating behaviors. In Chapter 4 we further investigated the effects of personality within a specific mating dyad. Recent studies suggest that mate compatibility within personality types can improve reproductive performance measurements such as mating success, offspring survivorship, and offspring quality. We investigated the effect of personality compatibility on reproductive performance through analyzing mate dyad similarity in personality (similar versus dissimilar), specific personality combinations, and relative personality scores (male scores higher or lower than female scores). The specific personality combinations were: females and males that were both low on the personality component (L_L), females were low but males were high (L_H), females were high but males were low (H_L), and females and males were both high (H_H). Our results suggest that giant pandas appear to be assortatively mating based on aggressive, fearful, and stereotypical-excitable personality traits. Mate dyads that were dissimilar on the stereotypical-excitable component score were more likely to have successful intromission than pairs that were similar. L_H mate dyads on the aggressive and stereotypical-excitable personality traits had increased intromission success and cub production than all other possible personality combinations. L_L on fearfulness had higher intromission success but not higher cub production than all other possible personality combinations. Mate dyads composed of males ranked high on the aggressive component relative to the female resulted in higher reproductive performance and cub production. In captive breeding programs, mates are traditionally selected solely on the basis of genetic parameters to minimize loss of genetic diversity and inbreeding coefficients. Our results suggest that integrating genetic with behavioral measures of preference, breeding history factors, and personality would increase the reproductive output of the giant panda captive-breeding program. We stress the importance of considering husbandry practices to accommodate personality traits and ultimately increase reproductive performance in the captive population while simultaneously maintaining both genetic and behavioral diversity. The relative influence of some significant life experiences on youths' response to environmental education, including environmental service-learning, was also considered. Telephone surveys were used to gather data from 22 Portland metropolitan area environmental education programs. Data included 2014 annual biophysical impacts (e.g., area of invasive species removed, pounds of litter removed) and information on programming (e.g., length of program, % time outside). Eleven programs administered a 33-question environmental literacy assessment to participants of their programs (n=393). The assessment included the New Environmental Paradigm, the Inclusion of Nature in Self, questions from Environmental Identity Scale and self-constructed questions. One 8th grade program was identified for a detailed case study. In this 8th grade programs, slight variations in educational activities occurred among three treatment groups which varied the amount of time youth spent engaged in ecomanagement. Youth from the three treatment groups and a control group were administered the environmental literacy assessment at the beginning and end of the program. Qualitative data for the youth in the treatment groups were gathered to further consider how environmental literacy was impacted by participation in the program. Despite holding great promise, the ability to determine and invest in only the most promising program alternatives plays a key role in the successful use of energy efficiency as a utility-wide resource. This issue becomes even more significant considering the availability of a vast number of potential energy efficiency programs, the rapidly changing business environment, and the existence of multiple stakeholders. This dissertation introduces hierarchical decision modeling as the framework for energy efficiency program planning in electric utilities. The model focuses on the assessment of emerging energy efficiency programs and proposes to bridge the gap between technology screening and cost/benefit evaluation practices. This approach is expected to identify emerging technology alternatives which have the highest potential to pass cost/benefit ratio testing procedures and contribute to the effectiveness of decision practices in energy efficiency program planning. The model also incorporates rank order analysis and sensitivity analysis for testing the robustness of results from different stakeholder perspectives and future uncertainties in an attempt to enable more informed decision-making practices. The model was applied to the case of 13 high priority emerging energy efficiency program alternatives identified in the Pacific Northwest, U.S.A. The results of this study reveal that energy savings potential is the most important program management consideration in selecting emerging energy efficiency programs. Market dissemination potential and program development and implementation potential are the second and third most important, whereas ancillary benefits potential is the least important program management consideration. The results imply that program value considerations, comprised of energy savings potential and ancillary benefits potential; and program feasibility considerations, comprised of program development and implementation potential and market dissemination potential, have almost equal impacts on assessment of emerging energy efficiency programs. Considering the overwhelming number of value-focused studies and the few feasibility-focused studies in the literature, this finding clearly shows that feasibility-focused studies are greatly understudied. Viral metagenomics offers a potential means of establishing a more comprehensive view of virus diversity and evolution, as vast amounts of new sequence data becomes available for comparative analysis.Metagenomic DNA from virus-sized particles (smaller than 0.2 microns in diameter) was isolated from approximately 20 liters of sediment obtained from Boiling Springs Lake (BSL) and sequenced. BSL is a large, acidic hot-spring (with a pH of 2.2, and temperatures ranging from 50°C to 96°C) located in Lassen Volcanic National Park, USA. BSL supports a purely microbial ecosystem comprised largely of Archaea and Bacteria, however, the lower temperature regions permit the growth of acid- and thermo-tolerant Eukarya. This distinctive feature of the BSL microbial ecosystem ensures that virus types infecting all domains of life will be present. The metagenomic sequence data was used to characterize the types of viruses present within the microbial ecosystem, to ascertain the extent of genetic diversity and novelty comprising the BSL virus assemblage, and to explore the genomic and structural modalities of virus evolution.Metagenomic surveys of natural virus assemblages, including the survey of BSL, have revealed that the diversity within the virosphere far exceeds what has currently been determined through the detailed study of viruses that are relevant to human health and agriculture. The number of as-yet-uncharacterized virus protein families present in the BSL assemblage was estimated by clustering analysis. Genomic context analysis of the predicted viral protein sequences in the BSL dataset indicates that most of the putative uncharacterized proteins are endemic or unique to BSL, and are largely harbored by known virus types. A comparative metagenomic analysis approach identified a set of conserved, yet uncharacterized BSL protein sequences that are commonly found in other similar and dissimilar environments.New sequence data from metagenomic surveys of natural virus assemblages was also used to better characterize and define known virus protein families, as some of the viruses found in the BSL environment represent distant relatives of well-characterized isolates. By comparing viral genes and protein sequences from these highly divergent species, it is possible to better understand the dynamics of adaptation and evolution in the virosphere. However, work-family interventions are rare and are generally not thoroughly evaluated. The current study seeks to better understand the impacts of STAR ("Support. Transform. Achieve. Results."), the large-scale work-family intervention developed and implemented by the Work, Family, & Health Network (see Bray et al., 2013). Drawing on Conservation of Resources theory (Hobfoll, 1989), this study examines supervisors' participation in STAR through assessment of three primary supervisor-specific outcomes: training-related views and behaviors, well-being, and the work-family interface. The sample, consisting of 184 supervisors from 30 extended-care facilities throughout the northeastern United States, comes from archival data that were collected by the Work, Family, & Health Network. Results show a lack of support for STAR intervention effects on supervisor-level outcomes. Despite the lack of statistically significant effects on supervisors, it is important to note the lack of iatrogenic effects, indicating that participation in the STAR intervention did not harm supervisor outcomes. Implications, future directions, and limitations of the study are discussed.
