The program's winter germplasm is quite susceptible to Barley Yellow Dwarf Virus (BYDV). The program recently released the spring 2-row variety Orca, which carries the Yd2 gene for BYDV resistance. The objective of this project was to transfer the Yd2 gene from Orca to the winter varieties Strider and Kold. A molecular marker tightly linked to the Yd2 locus was used to facilitate the introgression of this gene from the spring to the winter ger nplasm pool In 1998, six-row plants were selected in F2 populations derived from crosses of Orca x Kold and Orca x Spider. DNA was extracted from these plants for genotyping with the molecular marker YLM, which is tightly linked to the Yd2 locus on the long arm of chromosome 3 (311). In order to encourage natural infection by BYDV-infected aphids, the F3 families tracing to selected F2 plants were planted one month before the usual planting date in the fall of 1998. Unusually low temperatures in December 1998 led to significant winter injury, leading to the loss of approximately 70% of the F3 families in each of the two populations. Phenotype data (visual rating of BYDV symptoms on F2 plants and F3 families, and visual rating of winter survival in F3 families) and genotype data (allelic structure of each F2 plant at the YLM locus) were collected to determine the genotype of the F3 phenotypic selections and to assess the utility of the marker locus as tools for rapidly introgressing target alleles from spring to winter germplasm. Horticultural and nursery crops are the leading agricultural commodity in Oregon. Among currently grown nursery plants, Verbena x hybrida is a staple ornamental. It is grown and shipped worldwide, thus requiring a high level of certainty concerning the plants’ health and potential viability. V. x hybrida cultivars have been infected by a potyvirus that has been a sporadic but chronic problem in the floriculture industry for many years. Within the plant, detectable potyvirus titers fluctuate widely, making it difficult to detect in commercial nurseries. Virus detection can also be affected by changing environmental conditions, which also influence virus levels. The objectives of this study were to determine the environmental conditions that optimized detectable virus levels, to observe if the virus could be detected in tissue culture plantlets, to establish the earliest age at which plants could be accurately assayed, and to ascertain the most sensitive and reliable testing method for virus detection. The V. x hybrida cultivars in the study were infected ‘Lavender Shades’ and non-infected ‘Twilight with Blue Eye.’ Plants were tissue cultured or potted and placed in growth chambers at three temperatures with constant photoperiod and then at three photoperiods with constant temperature. The Enzyme-linked Immunosorbent Assay (ELISA) and Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) were used as the testing methods. Tests were performed at two week intervals for a duration of two months in each of the two growth chamber experiments. The resulting data support the conclusion that V. x hybrida plants should be grown in pots at 26°C and 16 or 20 hour photoperiod for at least 6 weeks before testing with RT-PCR for the most accurate detection of virus. Because global warming is predicted to increase the severity of El Niño events in the future, understanding how warmer temperatures affect seaweed growth and the animals associated with seaweeds is important for predicting how marine ecosystems will change. The Galapagos Islands of Ecuador are considered a living laboratory for studying climate change and served as the experimental site for this study. An artificially induced average increase in temperature by 1.45°C significantly decreased seaweed biomass in the intertidal of Tortuga Bay, Santa Cruz Island. The experiment utilized a block design with six treatments. Treatments consisted of 15cm x 15cm black (warm) and white (cool) plates to cause differential thermal stress, cross-factored with herbivore exclusion cages to study the effects of grazing, and a natural substratum control. Thermal stress (plate color) and grazing were both significant factors reducing seaweed biomass. Abundance, richness, and diversity of invertebrate animals living among the seaweeds showed declines in warmer treatments. Effects on small animals could reflect intolerance to thermal stress, loss of habitat (seaweed), or both, and future studies should investigate these factors. These results indicate that warmer conditions due to human induced climate change can decrease seaweed growth and affect animals that rely on seaweeds for food or habitat, possibly including the Galapagos marine iguana. Insight from the Galapagos might help predict seaweed response to climate change in other regions including the temperate Pacific Northwest.
