Information provided from population counts can give insight to soil ecologists about the effects of human land use, monitoring soil and water health, benefits of soil biodiversity in agriculture, and the impact of global climate change. The process of performing population counts, which includes classifying and sorting specimens, is very time consuming because of the large diversity and quantities of specimens in soil samples. A mechanical system was designed and developed to automate this process. The system features automated imaging, a pair of linear stages for automated positioning, and a six-axis robot with a pipette end effector for sorting. Images acquired with the system will be used by pattern recognition software to classify specimens from a soil sample. The final system was successful in creating high quality images of 19 species of soil mesofauna and sorting them. Madagascar, home to a high degree of endemic species and has been subject to numerous programs and organizations intent on preserving its biodiversity, which is currently pressured largely by anthropogenic causes. Protected areas, including national parks, make up a central part of these conservation efforts, combining ecotourism and restricting access for locals. This study looked to see how local people and conservation efforts interact, and how villagers view the concept of conservation at one site of one of Madagascar’s National Parks (Malio, at Andohahela National Park). In looking at viewpoints and opinions about conservation, park management and related factors, locals revealed a varying depth of knowledge about conservation and the systems supporting the park’s function. While the feelings of villagers regarding the park varied as well, it was largely considered problematic and many villagers expressed frustrations with limitations of the park impacting livelihoods, problems with park management, inadequate compensation and an inability to express their own needs regarding park planning and management. This localized study reflected challenges of a larger problem, coordinating the needs for livelihoods and conservation of Madagascar’s biodiversity, complicated by differential actions of interacting systems and organizations, cultural undercurrents, as well as systemic problems. However, little is known about the diversity and function of microorganisms in the atmosphere. To investigate the forces driving the assembly of bacterial microbial communities in the atmosphere, I measured temporal variation in bacterial diversity and composition over diurnal and inter-day time scales. Results suggest that bacterial communities in the atmosphere markedly vary over diurnal time scales and are likely structured by inputs from both local terrestrial and long-distance sources. To assess the potential functions of bacteria and fungi in the atmosphere, I characterized total and potentially active communities using both RNA- and DNA-based data. Results suggest there are metabolically active microorganisms in the atmosphere that may affect atmospheric functions including precipitation development and carbon cycling. This dissertation includes previously published and unpublished co-authored material.
