While many gene families follow predictable patterns of molecular tinkering, many protein families exist with completely novel functions now essential. The guanylate kinase protein interaction domain (GKPID) of the membrane associated guanylate kinases (MAGUK) represents a model system for the study of protein evolution in which a protein scaffolding domain has evolved from a nucleotide kinase ancestor. Here we elucidate the ancient mechanisms by which these new functions evolved by combining ancestral protein reconstruction with in vitro and cell-biological molecular experiments. We found that the GKPID's capacity to serve as a mitotic spindle-orienting scaffold evolved by duplication and divergence of an ancient guanylate kinase enzyme before the divergence of animals and choanoflagellates. Re-introducing a single historical substitution into the ancestral guanylate kinase is sufficient to abolish the ancestral enzyme activity, confer the derived scaffolding function, and establish the capacity to mediate spindle orientation in cultured cells. This substitution appears to have revealed a latent protein-binding site, rather than constructing a novel interaction interface, apparently by altering the dynamics or conformational occupancy of a hinge region that determines whether the binding site is exposed or hidden. Three further substitutions also conveyed a measure of ligand specificity to phosphorylated Pins, which is necessary in metazoan spindle orientation pathways. These findings show how a small number of simple, ancient genetic changes caused the evolution of novel molecular functions crucial for the evolution of complex animals and laid the groundwork for an entirely new family of metazoan scaffolding proteins.   Citation × Citation Citation Abstract Copy Title: Synthesis and Characterization of Rotationally Disordered Intergrowths Containing PbX and TiX2 (X = Se and Te) Author: Moore, Daniel Year: 2015 The method of modulated elemental reactants (MER) is used to prepare the layered compound Ti1+xSe2. Using a thin-film precursor prepared by sequentially depositing elemental titanium and selenium by physical vapor deposition, the binary compound is readily formed by short-duration annealing at 350°C. The structural and electrical properties of this compound are investigated and suggest that the film is markedly different from the bulk crystalline material. The synthesis is then expanded to produce the first TiSe2 turbostratically disordered misfit layer compound, (PbSe)1.16TiSe2. With the synthesis technique established, the second compound in this family, (PbSe)1.18(TiSe2)2, was synthesized and compared to the misfit layer compound synthesized by bulk methods. The rotationally disordered compound demonstrated an increased Seebeck coefficient and reduced resistivity at room temperature, sparking interest in this family of compounds as potential thermoelectric materials. Designed synthesis of several targeted compounds is reported with interest in evaluating thermoelectric potential of these rotationally disordered compounds. The first nine members of this family were synthesized with m and n values 1 - 3 and evaluated for their thermoelectric properties. Results from this study indicated compounds with larger n values could be of further interest and many more of these compounds are synthesized and reported. Designed synthesis using the MER synthesis method is then pushed further to synthesize the first telluride misfit layer compounds. Four new compounds of the [(PbTe)1.17]m(TiTe2)n family are reported, and their structures were examined by a variety of X-ray diffraction techniques. The robustness of the MER method is revealed by the synthesis of these layered intergrowths which would not be possible using other synthesis methods available. An increasing amount of attention is being directed to parental mental health and addressing the well-being of the entire family system. In order to best serve families raising children with ASDs, the present study sought to better understand the relation between social support, various risk factors, and caregiver depression. Data were collected from 60 families with children 2—7 years with ASDs through the use of extensive, in-home interviews with primary caregivers. Mothers reported the availability and helpfulness of both formal and informal supports. Similar to previous research, mothers also reported elevated levels of depression. In the present sample, more than half of mothers reported depressive symptoms at or above the cut-off for mild depressive symptomatology. Both child-related variables (autism symptomatology, atypical behavior) and service-related variables (satisfaction with the education eligibility process, satisfaction with sources of information about ASDs) were predictive of maternal depression. Social support and maternal depression were not related.
